Using a USB feed from my MacBook of a lossless audio file, I prefer the sound via the Oppo. I couldn't say if it's the DAC or if it's the amplifier driving the headphones, but it sounds more musical to my ears. But they both sound pretty decent and I never had any complaints (still don't) about the Presonus before I bought the Oppo.
However there is another option if your audio interface has a digital output, and that is the TC BMC2. Very nice DAC in there and drives headphones well. Usually around a ton secondhand.
MIDI instruments don't make their own sounds. You need to send the MIDI into a computer that has software instruments which convert those commands into sounds. Line in is for analogue audio signals, not digital commands. (Unless I've misunderstood you).
The Presonus also has MIDI in/thru and MIDI out 5 pin sockets. It's one of the reasons I bought it over a Focusrite (which did not have them). At that time, my old keyboard didn't have USB MIDI. So, I plugged it into the Presonus and it just worked.
FWIW, the other reason was I often use my iPad as a DAW for tracking and the Presonus supported that.
